New vector enhances
protein expression
pOET3 combines the flexibility and convenience
offered by the pOET1 and pOET2 vectors, with enhanced
capabilities that complement the improvements in protein
expression delivered by the Company’s successful
flashBAC vectors. pOET1 and pOET2 are
baculovirus transfer plasmids designed for high-level
expression of foreign genes under the powerful AcMNPV
polyhedrin (polh) promoter.
pOET3 protein expression is driven from the AcMNPV p6.9
promoter, enabling earlier expression compared with the polh
promoter used in pOET1 and pOET2. The recombinant protein is therefore
produced at a time during the infection process when the cellular
machinery is in a much healthier condition, compared to late times in
infection. This can give significant improvements when proteins that
require extensive post-translational modifications are produced, such as
glycoproteins destined for secretion or membrane insertion.
pOET3 plasmids are also:
- Compatible with all baculovirus systems based on homologous
recombination.
- Have the same MCS and small size as pOET1 to facilitate easy
cloning.
The release of pOET3 follows Oxford Expression Technologies’ launch
of its successful suite of transfer plasmids with unique signal
sequences and promoters, including pOET1 and pOET1N that increase
cloning efficiency and expression of foreign genes.
